Both qualitative and quantitative analysis can be performed using real-time digital pcr. A variety of cell and gene therapy applications, including viral titer and vector copy number measurements, gene and car t-cell therapy development and manufacturing, are made possible by digital pcr applications. While pcr involves enzymatic amplification of dna, rt-pcr involves starting the process from scratch with rna. The rt-pcr technique differs in that it includes an additional step of reverse transcription of rna to dna before beginning the amplification step. The sample is partitioned into 20,000 droplets of a defined volume in ddpcr, and the rna or dna is distributed randomly among the droplets. The droplet reader is sensitive enough after amplification to detect the fluorescence signal in each individual droplet. This is how digital pcr works: a bulk pcr reaction is divided into thousands of nanoliter-scale reactions, each containing zero, one, or a few dna molecules. The sample is absolute quantified by counting positive reactions and using poisson statistics. Real-time pcr collects data during the exponential growth phase of pcr, whereas traditional pcr collects data at the reaction's end-point (end-point pcr vs real-time). The digital pcr principle is used to achieve absolute quantification by partitioning the reaction. The conventional pcr can only precisely amplify dna up to 2000 nucleotides, whereas the rtpcr or qpcr can amplify dna as well as quantify the amount of dna (pcr vs qpcr).
